Clostridium perfringens toksinlerinin tesbitinde fare toksisite testlerine alternatif olarak ELISA 'nın uygulanması

Özet
Bağırsak içeriğinde, kültürlerde ve buffer solüsyonunda CI. perfringens beta ve epsilon toksinleri tesbit etmek amacıyla double sandwich ELISA uygulandı. Bu amaçla mikroplaklar spesifik koyun antitoksik serumlar ile kaplandı ve tavşanlarda hazırlanan spesifik antitoksik serumlar dedekte edici olarak kullanıldı. Double sandwich ELISA ile bağırsak içeriğinde minimum 15.6 ng/ml beta ve 7.8 ng/ml epsilon toksin; buffer solüsyonunda ise minimum 3.9 ng/ml beta ve 1.9 ng/ml epsilon toksin tesbit edildi. Aynı şartarda yapılan beta ve epsilon ELISA tekrarlarında intra test varyasyon katsayıları % 10'un altında bulunmuştur. Farklı seviyede toksin içeren kültürlerde fare toksisite testleri ile double sandwich ELİSA değerleri uyum gösterdi. Yüksek seviyede toksin içeren kültürlere ait ELISA OD değerleri, orta ve düşük seviyede toksin içerenlere göre yüksek değerler verdi. Toksin içeren tüm kültürler, içermeyenlere göre belirgin olarak yüksek OD değerleri verdi. Bulgulara göre double sandwich ELISA'nın CI. perfringens beta ve epsilon toksinleri tesbit edebildiği ve fare toksisite testlerine alternatif olarak kullanılabileceği sonucuna varılmıştır.
Abstract
Specific double-sandwich ELISAs for beta and epsilon toxins of CI. perfringens in buffers, culture supernatants or in intestinal contents, were developed by coating wells of microplates with specific sheep antitoxin and using specific rabbit antitoxin as detecting antibodies. When applied to detect the toxin in intestinal contents, amounts were about a minimum level of 15.6 ng/ml for beta and 7.8 ng/ml for epsilon toxins; in buffer a minimum level of 3.9 ng/ml for beta and 1.9 ng/ml for epsilon toxins. When the same intestinal contents were retested by Double Sandwich ELISA with the same equipments and the same preparative lots of reagents for beta and epsilon toxins, the intra assay coefficent of variation in each assay were less than 10 %. There were conformity between mouse toxicity assay and double sandwich ELISA in which containing different levels of toxins in cultures. ELISA OD values belonging to the culture contained high levels of toxin, were higher than the others which contain medium or low levels of toxins. All cultures contained toxins, absolutely gave high OD values according to the media not to contain toxins. These results suggested that double sandwich ELISA described in this study could detect beta and epsilon toxins of CI. perfringens and could be used instead of mouse toxicity assay as alternatively.
