Özet
Beta talasemi majorlü (BTM) ve demir eksikliği anemili (DEA) çocuklarda lipid peroksidasyonunu ve antioksidan enzim aktivitelerini incelemek amacı ile bu çalışma planlandı. Çalışmaya, 20'er BTM ve DEA'lı hasta alındı. BTM'li hastalar düzenli olarak şelasyon tedavisi ve 3 haftada bir eritrosit transfüzyonu alıyorlardı. Kontrol grubu olarak da, hematolojik bulguları normal olan, yaş ve cinsiyet bakımından uyumlu 20 sağlıklı çocuk seçildi. Hasta ve kontrol gruplarında malondialdehit (MDA) düzeyi, süperoksid dismutaz (SOD), glutatyon peroksidaz (GSH-Px) ve katalaz aktiviteleri ölçüldü. Hasta gruplarının değerleri birbiriyle ve kontrol grubu ile karşılaştırıldı. MDA seviyesi; BTM grubunda, DEA ve kontrol grubuna göre; DEA grubunda ise, kontrol grubuna göre anlamlı olarak yüksekti (p<0.05, p<0.05). SOD ve katalaz aktiviteleri bakımından hasta ve kontrol grupları arasında anlamlı fark yoktu. GSH-Px aktivitesi; BTM grubunda, DEA ve kontrol gruplarına göre anlamlı olarak düşüktü (p<0.05, p<0.05). Buna gör e, her iki hasta grubunda eritrosit lipid peroksidasyonun, kontrol grubuna göre anlamlı olarak arttığı, bu artışın BTM'de daha fazla olduğu, bunun da anemi patojenezinde rol oynayabileceği sonucuna varıldı. Ayrıca değişik yollarla oluşabilecek oksidatif stresin monitörizasyonu için serum MDA seviyesinin güvenilir bir marker olarak kullanılabileceği düşünüldü.
Abstract
This study was planned to examine the lipid peroxidation and the antioxidant enzymes activities in children with iron deficiency anemia (IDA) and beta thalassemia major (BTM). Twenty patients with IDA and twenty patients with BTM were enrolled in this study. Twenty healthy children whose hematological findings in normal ranges were selected as control group. They were also age and sex matched. Serum levels of malondialdehyde (MDA) and activities of superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), catalase were measured in all groups. The results were compared to each other and those of healthy subjects. In conclusion, serum MDA level was increased significiantly in BTM group compared to the IDA and the controls (p<0.05, p<0.05). In the IDA group, MDA level was higher than that of control group (p<0.05). SOD and catalase activities were not significantly different in all groups. GSH-Px activity in BTM was significantly lower than those of both the IDA group and the controls (p<0.05, p>0.05). Accord ing to these results ıt was suggested that susceptibility of RBCs to lipid peroxidation which may be play a role in the pathogenesis of the anemia in both IDA and BTM were increased. It was thought that serum MDA level could be use as a marker in the monitoring of the oxidant stress in IDA and BTM.
