Özet
AMAÇ: Apoptoz, immün sistemin gelişimi, regulasyonu ve fonksiyonları için önemli role sahip fizyolojik bir ölüm şeklidir. Fas (CD95), apoptozun düzenlenmesinde anahtar rol oynayan, timositlerde ve aktif T lenfositlerinde bulunan TNF reseptör ailesinden bir yüzey reseptörüdür. SLE'de DNA ve histonların da içinde bulunduğu çok sayıda nükleer antijene karşı antikor oluşumu söz konusudur ve hücre dışı nükleer antijenlerin kaynağının artmış apoptoz olabileceği düşünülmüştür. In vitro olarak hastalık aktivitesi ve apoptozun hızı arasında belirgin korelasyon olduğu ve hücre dışına salınan bu nükleer antijenlerin immün cevap ve immün kompleks oluşumuna neden olduğu öne sürülmüştür. Fas ile ilişkili apoptozda reseptör-ligand ilişkisi ile oluşan hücre ölümü söz konusudur. Efektör hücrelerde bulunan FasL hedef hücredeki Fas ile ilişkili hedef hücredeki apoptotik sinyallerin aktivasyonuna neden olur. Fas ve FasL periferik toleransın sağlanmasında ve T hücre gelişiminde rol alırlar. Bu çalışmada SLE'li hastaların periferik kan örneklerinde Fas antijen ekspresyonunu hem naive hem de hafıza CD4+ ve CD8+ T lenfositlerde göstermeye çalıştık. Ayrıca Fas ekspresyonunun hastalık aktivitesi ile ilişkili olup olmadığını da araştırdık. GEREÇ VE YÖNTEM: 12 aktif SLE'li hastadan ve 25 inaktif SLE'li hastadan kan örnekleri alındı. 29 sıhhatli kişilerden normal kan örnekleri alındı. Fas (CD95) ekspresyonu akım sitometrisi ile belirlendi. BULGULAR: Elde ettiğimiz sonuçlarda; CD4+ T hücre populasyonunda hafıza (CD45RO+) hücrelerde ve naive hücrelerde Fas antijen ekspresyonunu kontrol grubuna göre anlamlı olarak yüksek bulduk ( p 0.05). Hastalık aktivitesi ile Fas ekspresyonu arasında ise anlamlı bir ilişki tespit edilmedi. SONUÇ : Apoptozdaki anormalitelerin, SLE patogenezinde katkısı olabilir
Abstract
OBJECTIVE: Apoptosis is a physiological way of death which has an important role in the evolution, regulation and functions of the immune system. Fas (CD95), a surface antigen from the TNF receptor family is found in thymocytes and active T Iymphocytes and plays a key role in the regulation of apoptosis. In SLE, there is the regulation of apoptosis against a lot of nuclear antigens, induding DNA and histones, and it is thought that the source of the extracellular nuclear antigens may be apoptosis. It is claimed that in vitro there is a distinct correlation between disease activity and apoptosis rate, and this nuclear antigens excreted from the cells cause immun response and immun complex formation. In Fas related apoptosis there is a cellular death caused by the receptor ligand relation. FasL found in effector cells, activates the apoptotic signals in Fas related target cells. Fas and FasL plays a role in the maintainance of peripheral tolerance and T cell growth. In this study we tried to show the Fas antigen expression in both naive and memory CD4 + or CD 8 + T lymphocytes taken from the peripheral blood samples of SLE patients. We also investigated the probability if Fas expression had a relation with disease activity. MATERIAL AND METHODS: Blood samples was obtained from 12 patients with active SLE and from 25 patients with inactive SLE. Normal blood samples was obtained from 29 healthy volunteers. Fas (CD95) expression was confirmed by flow cytometry. RESULTS: According to our study results we found that in CD4+ T cell population, the Fas gene expression was significantly higher in both memory (CD45RO+) and naive cells compared with the control group (p<0.05). In CD 8 + T cell population the Fas gene expression in the memory cells was significantly higher than the control group (p<0.05), while no difference was found between the control group and naive cells ( p>0.05). In addition, no significant correlation was found between disease activity and Fas expression. CONCLUSION : These abnormalities of apoptosis may contribute to pathogenesis of SLE.
