Koyunlarda B. melitensis'in mikroaglütinasyon testi (MAT) ile teshisi üzerinde arastirmalar

Özet
Bu çalışmada Brucella melitensis enfeksiyonuna karşı serumda şekillenen antikorlar Plate Test (PT), Rose Bengal Plate Test (RBPT), Tüp Aglütinasyon Testi (TAT) ve Mikroaglütinasyon Testi (MAT) ile saptanarak sonuçlar sensitiflik ve duyarlık açısından karşılaştırıldı. MAT'de B. melitensıs'ten hazırlanan ve Safranin-0 ile boyalı antijen kullanıldı ve test U tabanlı mikropleytlerde gerçekleştirildi. Teste tabi tutulan 1580 adet koyun kan serumundan PT'de 341 (%21.6). RBPT'de 358 (%22.7), TAT'de 463 (%29.3) ve MAT'de 587 (%37.1) adet pozitif reaksiyon şaptandı. Mikroaglütinasyon Testi'nin brucella antikor fitrelerinin belirlenmesi için daha az zaman, kısa inkübasyon süresi ve daha az antijen kullanımı gerektirdiğinden avantajlı olduğu saptandı. Koyunlarda B. melitensis enfeksiyonunun serolojik tanısında diğer testlerin yanı sıra MAT'nin kullanılmasının enfeksiyonun saptanmasında yararlı olacağı sonucuna varıldı.
Abstract
In this study, Plate Test (PT), Rose Bengal Test (RBPT), Tube Agglutination Test (TAT) and Microagglutination Test (MAT) were used for the detection of Brucella melitensis antibodies in sheep sera. Each results of these tests were compared wjth other tests. The antigen which prepared by B. melitensis and stained Safranin-0 was used to microagglutination Test (MAT) and the test was confirmed in the U bottom microplates. Among 1580 blood serum samples examined and 341 (21.6%), 358 (22.7%), 463 (29.3%), 587 (37.1%) samples were determined as positive in PT, RBPT, TAT and MAT respectively. Advantages of the microagglutination test were that it required less time to perform, had a shorter incubation period, and uses less antigen. In detecting B. melitensis infection in sheep the use of MAT, together with other tests was found to be useful.
