Cystic echinococcosis (CE)' in serolojik tanısında yalancı pozitifliklerin araştırılması

Investigation of false positive reactions in sero-diagnosis of cystic echinococcosis

Özet

Çalışmaya Cystic echinococccosis (CE) serolojisinde yalancı pozitifliği araştırmak amacıyla; basit kist, tümör, apse vb. hastalığı olan 71 olgu, CE olduğu operasyonla kanıtlanmış 35 olgu ve 30 sağlıklı insan alınmıştır. Olgulardan alınan serumlarda İndirekt Fluoresan Antikor (IFA), İndirekt Hemaglutinasyon(IHA), Enzyme-Linked Immunosorbent Assay (ELISA) ve Sodium Dodecyl Sulfate Polyacrylamide Gel Electrophoresis (SDS-PAGE) ve Western blot (Immunoblotting) yöntemleri ile spesifik antikorlar araştırılmıştır. Basit kist, tümör, apse vb. hastalığı olanların oluşturduğu 71 kişilik grupta IFA ile % 12.7, IHA ile %9.9, ELISA ile %11.3 oranında antikor tespit edilirken; 30 sağlıklı insan serumunda IFA ve ELISA yöntemleri ile %2.9 oranında yalancı pozitiflik saptanmıştır. WB yöntemi ile her iki grupta da herhangi bir yalancı pozitiflik elde edilmemiştir. CE olduğu operasyonla kanıtlanmış 35 hastada IFA ile %91.4, IHA ile %97.1, ELISA ile %100, SDS-PAGE ve WB ile %94.3 oranında seropozitiflik saptanmıştır

Abstract

In this study, 71 sera from patients with nonhydatid disease (simple cyst, tumour, abscess etc.); 35 sera from patients with cystic echinococcosis, confirmed by surgery and 30 sera from healthy people were obtained. Specific antibodies were investigated in these serum samples by Indirect Fluorescent Antibody (IFA), Indirect Hemagglutination (IHA), Enzyme Linked Immunosorbent Assay (ELISA), Sodium Dodecyl Sulphate Polyacrylamide Gel Electrophoresis (SDS-PAGE) and Western blot (WB) tests. While specific antibodies were found in the group of nonhydatid disease was consisted of 71 patients by IFA, IHA and ELISA tests 12.7%, 9.9% and 11.3% respectively, false positive reactions were obtained in the sera of healthy people by IFA and ELISA (2.9%) tests. Of sera from patients with nonhydatid disease and healthy people, none were positive by Western blotting. 35 CE patients confirmed by surgery were found positive by IFA (91.4%), IHA (97.1 %), ELISA (100%), SDS-PAGE and WB (94.3%) tests
