Nikel Klorürün Küçük Hücreli Olmayan Akciğer Kanseri Hücre Dizilerinde (A549) Hücre Canlılığı ve Koloni Formasyonu Üzerine Olan Etkisi

Özet

Amaç: Son yıllarda birçok metal ve metal bileşiğinin kanser tedavisinde umut vadedebileceği gösterilmiştir. Bu elementlerden bir tanesi de nikeldir. Her ne kadar nikel bir genotoksik karsinojen olarak tanımlansa da bazı kanser türlerinde terapötik bir ajan olabileceği ileri sürülmüştür. Ancak nikelin küçük hücreli olmayan akciğer kanseri hücre dizilerinde (A549) hücre proliferasyonu ve kanser hücresi koloni formasyonu üzerine olan etkileri bilinmemektedir. Yapmış olduğumuz çalışma ile nikel klorürün (NiCl2 ) A549 hücre dizilerinin hücre canlılığı ve koloni oluşturma yetenekleri üzerine olan etkilerinin araştırılmasını amaçladık. Yöntem: NiCl2 ’ün hücre canlılığına olan etkilerinin belirlenmesi için A549 hücre dizilerine 50 / 100 / 200 / 400 / 800 / 1600 / 3200 µM dozlarında 24, 48 ve 72. saatlerde NiCl2 uygulaması yapıldı. Hücre canlılığına olan etkilerinin belirlenmesi için MTT (3-(4,5-dimethylthiazol-2-yl)-2,5- diphenyltetrazolium bromide) analizi gerçekleştirildi. Koloni sayılarına olan etkilerinin belirlenmesi için ise koloni formasyon analizi uygulandı. Bulgular: NiCl2 ’ün 24. saatte 100 µM’dan itibaren (p<0.01), 48 ve 72. saatlerde 50 µM’dan itibaren (p<0.01), hücre canlılığını istatistiksel olarak anlamlı bir şekilde azalttığı görüldü. NiCl2 ’ün A549 hücrelerinin büyümesini %50 oranında baskılayan dozunun (IC50) ise 24, 48 ve 72. saatlerde sırasıyla 535,7 µM, 322,7 µM ve 215,9 µM olduğu hesaplandı. Ayrıca, IC50 dozunda NiCl2 uygulanan A549 hücre dizilerinde oluşan koloni sayısının (koloni sayısı: 34,2 ± 6,02), kontrol grubuna göre (koloni sayısı: 67,3 ±5,81) istatistiksek olarak daha az olduğu belirlendi (p<0.01). Sonuç: Bulgularımız, küçük hücreli olmayan akciğer kanseri hücre dizilerine (A549) NiCl2 uygulamasının, doz ve zaman bağımlı olarak hücre canlılığını baskıladığı ve kanser hücrelerinin koloni yapabilme yeteneğini azalttığını göstermektedir.

Abstract

Introduction: Recently years, it has been shown that many metal and metal compounds might be a promising in cancer treatment. One of these elements is nickel. Although nickel is defined as a genotoxic carcinogen, it has been suggested that it may be a therapeutic agent in some types of cancer. However, the effects of nickel are unknown on cell proliferation and cancer cell colony formation in non-small cell lung cancer cell lines (A549). In our study, we aimed that investigate the effect of nickel chloride on cell viability and colony formation ability of A549 cell lines. Methods: In order to determine the effects of nickel chloride on cell viability, nickel chloride was treated to A549 cell lines for 24, 48 and 72 hours at 50 / 100 / 200 / 400 / 800 / 1600 / 3200 µM doses. MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) assay was used for determine of effect of nickel chloride on cell viability. Colony formation assay was used for determine of effect of nickel on colony number. Result: It was shown that nickel chloride decreased cell viability starting from 100 µM at 24th hour (p <0.01), starting from 50 µM at 48th and 72nd hour (p <0.01) statistically significantly. The IC50 (the half maximal inhibitory concentration) value was calculated as 535,7 µM, 322,7 µM and 215,9 µM doses at 24, 48, 72nd hour, respectively. Also, it was determined that the number of colonies (number of colonies: 34.2 ± 6.02) formed in A549 cell lines treated with Nickel chloride at IC50 dose was statistically less than the colony number of control group (number of colonies: 67.3 ± 5.81) (p <0.01). Conclusion: Our results indicate that the treatment of nickel chloride in non-small cell lung cancer cell lines (A549) decreases the colony formation ability of cancer cells and suppresses cell viability in a time and dose dependent manner.
