Doksorubisin uygulanmasının sıçan böbrek dokusunda meydana getirdiği değişiklikler üzerine benfotiamin'in koruyucu etkileri

Özet

Amaç : Doksorubisin bir antrasiklindir ve çeşitli hayvan modeli ve insan çalışmalarında benzer nefrotoksik etkilere sahip olduğu g österilmiştir. Bu çalışmada, doksorubisin uygulanan sıçanların böbrek d okularında meydana gelen değişiklikler üzerine benfotiaminin etkilerinin incelenmesi amaçlanmıştır. Gereç ve Yöntem: Çalışmada, 24 adet Wistar albino cinsi erkek sıçanlar kullanıldı. Deney hayvanları 4 eşit gruba ayrıldı. Kontrol grubuna dene y süresi olan 14 gün boyunca hiçbir uygulama yapılmadı. Doksorubisin grubuna doksorubisin 10 mg/kg intraperitoneal (i.p) tek doz uygulandı . Doksorubisin + Benfotiamin grubuna 10 mg/kg i.p tek doz doksorubisin verilmesinden sonra 70 mg/kg /gün dozunda benfotiamin oral o larak verildi. Benfotiamin grubuna ise 70 mg/kg /gün dozunda benfotiamin oral olarak uygulandı. Deney sonunda sıçanlar dekapite edildi ve böbrek dokuları çıkartıldı. Rutin takip işlemi ile dokular parafin bloklara gömüldü. Bloklardan alınan kesitlere Bax v e kaspaz -3 immünreaktivitesi için avidin -biotin- peroksidaz yöntemi uygulandı. Malondialdehit (MDA) seviyesi ise spektrofotometrik olarak belirlendi. Bulgular: Bax ve kaspaz -3 immünreaktivitesi kontrol ve benfotiamin gruplarında +1 yaygınlığında gözlendi. Kontrol gruplarıyla karşılaştırıldığında Doksorubisin grubunda Bax ve kaspaz -3 immünreaktivitesinde artış izlendi ve +3 yaygınlığında değerlendirildi. Doksorubisin grubu ile karşılaştırıldığında Doksorubisin + Benfotiamin grubunda Bax ve kaspaz -3 immünrea ktivitesinde azalma gözlendi ve +2 yaygınlığında değerlendirildi. MDA düzeyi Doksorubisin verilen grupta, kontrol grubu ve benfotiamin verilen grupla kıyaslandığında böbrek dokularında anlaml ı olarak artmış bulundu. Doksorubisin grubu ile karşılaştırıldığı nda Doksorubisin + benfotiamin verilen grupta MDA düzeyi anlamlı olarak azalmış bulundu. Sonuç: Bu çalışma doksorubisinin böbrek dokularında apoptozisi indüklediğini ve benfotiaminin doksorubisinin etkilerine karşı anti -apoptotik ve antioksidan etkileri olduğunu göstermiştir.

Abstract

Objective: Doxorubicin is an antracyc line that have similar nephrotoxic effects in both animal and human studies. In this study, we aimed to investigate the effects of benfotiamine on kidney tissues of the rats treated with doxorubicin. Material and Method: 24 wistar albino male rats were used in this study. Animals were divided to 4 equel groups. During the 14 day experiment period, control group did not receive any medication. 10 mg/kg intraperitoneal (i.p) doxorubicin was administered to Doxorubi cin group. In Doxorubicin+benfotiamine gro up; in addition to 10 mg/kg i.p single dose doxorubicin, 70 mg/kg benfotiamine was administered. Benfotiamine group received 70 mg/kg benfotiamine. At the end of the experiment, rats were decapitated and kidney tissues were removed. Tissues were embedded into parafine blocks with routine follow- up. Avidin- biotin -peroxidase method was applied to the sections obtained from bloks for Bax and caspase -3 immunoreactivity. Malondialdehyde (MDA) levels were measured by spectrophometer. Results: Bax and caspase -3 immunoreactivities were in +1 prevalence in control and benfotiamine groups. In doxorubicin group, bax and caspase - 3 immunoreactivity was increased in +3 prevalence in comparison with control group. In doxorubicin + benfotiamine group, -- bax and caspase -3 immunoreactivity was decreased to +2 prevalence in comparison with Doxorubicin group. -- In doxorubicin group,kidney tissue MDA levels were increased significantly in comparison with control and benfotiamine groups. MDA levels of Doxorubicin+benfotiamine gr oup decreased significantly in comparison with Doxorubicin group. Conclusion: In this study we observed that doxorubicin induces apoptosis in kidney tissues and benfotiamine have anti -apoptotic and antioxidant effec ts against Doxorubicin effects.
