Özet

Sepsis, yoğun bakım ünitelerinde mortalitenin en önemli nedenlerindendir. Nötrofiller, bakteriyel ajanlara karşı doğal immün savunmada önemlidir ve sepsis patogenezinde rol oynayan hücrelerdir. Bu çalışmada sepsisli hastalarda nötrofil fonksiyonlarında gözlenen değişikliklerin belirlenmesi amaçlanmıştır. Bu amaçla sepsis tanısı almış 10 hastaya ait nötrofillerin fagositik aktivitesi ve oksidatif patlama kapasiteleri araştırılmış ve elde edilen sonuçlar sağlıklı kontrollere ait nötrofil örnekleri ile kıyaslanmıştır. Fagositik aktivite ve oksidatif patlama kapasiteleri flow sitometri yöntemi ile değerlendirilmiştir. Fagositik aktivite, sepsisli hastalarda sağlıklı kontrollere göre azalmış olarak tespit edilirken, nötrofillerde oksidatif patlama kapasitesi sepsisli hastalarda artmış olarak bulunmuştur. Çalışmamızın sonuçlarına göre, sepsisli hastalarda nötrofil fonksiyonlarında bir bozulma olmaktadır ve bu durumun sepsisli hastalarda prognozu ve tedaviyi etkileyebileceği düşünülmüştür.

Abstract

Sepsis is one of the leading causes of mortality in intensive care unit. Neutrophils play an important role in the innate immun defense against bacterial agents and sepsis pathogenesis. The aim of this study was to evaluate the changes of neutrophil functions in patients with sepsis. Therefore, we evaluated neutrophil phagocytic activity and oxidative burst capacity in 10 patients with sepsis and compared the results with those of healthy control group. Phagocytic activity and oxidative burst capacity were assessed by flow cytometry. Neutrophil phagocytic activity was decreased in patients with sepsis compared to control groups. Whereas oxidative burst capacity was increased in patients with sepsis. These results indicate that there is an impairment in neutrophil function in patients with sepsis and it is considered that this condition may affect the prognosis and treatment of patients with sepsis.
