Özet

Bu araştırmanın amacı: (i) tüylülüğûn kalıtımının susam (Sesamum indicum L.) gen kaynağı materyalleri, TR 38106-1, TR 42804-1, TR 50235-1 ile belirlenmesi, (ii) üzerinde çalışılan materyaldeki tüylülüğû kontrol eden genlerin allelik ilişkilerinin ortaya konması ve (iii) tüylülük genlerinin direkt olarak gen kaynağı materyallerinden kombine edilmesi olmuştur. Kalıtım çalışması Fj, resiprokal Ft, ve BCJFJ jenerasyonları ile ortaya konmuştur. Fideler tüylü (T) ve tüysüz (TS) olarak sınıflandırılarak, açılmalar Khi-kare testi uygulanarak değerlendirilmiştir. Fj ve resiprokal Ft generasyonlannın 1 T: 0 TS segregasyonu TR 38106-1, TR 42804-1, TR 50235-1' daki genlerin kalıtımının dominant olduğunu ortaya koymaktadır. Fs ve resiprokal F[ genĞrasyonlannda farklılık olmaması tüylülük üzerinde maternal ya da sitöplazmik etkinin olmadığım ortaya koymaktadır. BCJFJ generasyonlannın 1 T: 1 TS ve F2 generasyonunda ailelerin 3T : 1 TS olarak açılma göstermeleri TR 38106-1, TR 42804-1 ve TR 50235-1' deki tüylülüğûn tek dominant gen tarafından kontrol edildiğini desteklemektedir. Allelizim için tüylü olarak belirlenen Fj bitkileri diallel olarak melezlenerek, diallel melezlerin test edilebilmesi amacıyla kendilenmişlerdir. Diallel melezlerin kendileme generasyonunda ailelerin 3T: 1 TS ve 15T : ITS olarak açılma göstermeleri TR 38106-1, TR 42804-1 ve TR 50235-1' deki tüylülük genlerinin allelik olmadığını ve üç ayrı gen olduğunu ortaya koymaktadır. Bu çalışma ile ilk olarak yerel çeşitler kullanılarak susamda (Sesamum indicum L.) tüylülüğûn kalıtımı araştırılmıştır.

Abstract

The objectives of this siudy were to: (i) determine inheritance of hairiness of sesame in the germplasm (Sesamum indicum L.) accessions (land races), TR 38106-1, TR 42804-1, TR 50235-1; (ii) study allelic relationships of genes among the accessions, and (iii) incorporate hairiness genes directly from the selected germplasm accessions. Inheritance of hairiness of sesame, was studied in Flt reciprocal Fj, and the BCjFi generations. Seedlings were classified as hairy (H) or non-hairy (N-H) and segregation ratios analyzed utilizing a Chi-square test for goodness of fit. Segregation ratios of one hairy (H) to zero non-hairy (N-H) seedlings in Fj and reciprocal Fj crosses indicated that dominant gene was controlling, hairiness in the selected plants of the accessions. There was no reciprocal differences found on hairiness indicating no cytoplasmic or maternal effect accounted for hairiness. The result of one H to one N-H segregation from backcross families confirmed that the hairiness was inherited as a single dominant gene. F; plants of accessions were selfed to obtain F2 generation. F2 seedlings were evaluated for hairyness. The observed three H to one N-H segregation ratio were in agreement with the results of diallel crosses and these segregation ratios were also in agreement with the original indication that a single dominant gene conditioning for hairyness that was derived from each of the sources. For allelism studies, dominant segregates of half-diallelcrosses among hairy Fj plants were selfed. Progeny segregations ofselfed diallel crosses resulted in three H to one N-H and fifteen H to one N-H ratios suggested that the genes for hairiness found TR 38106-1, TR 42804-1, and TR 50235-1 were not allelic and also were different from each other. This is the first study in which land races of Sesamum indicum L. accessions were directly utilized for inheritance studies of hairiness in sesame.
