Özet

Bitkisel materyallerden biyoaktif bileşiklerin ekstraksiyonu besinsel destek üretimi ve endüstriyel atıklardan bileşiklerin geri kazanımı gibi proseslerin en önemli aşamalarından biridir. Ekstraksiyon, solvent çeşidi, partikül boyutu, ekstraksiyon süresi ve sıcaklığı gibi faktörlerden etkilenmektedir. Bu araştırmada farklı çözgenlerle ekstrakte edilen alıç (Crataegus orientalis) meyvesinin toplam fenolik madde içeriğinin, antioksidan aktivitesinin ve fenolik madde profilinin belirlenmesi ve en uygun çözgenin tespiti amaçlanmıştır. Beyşehir’de doğal olarak yetişen ağaçlardan toplanan alıç meyveleri su, metanol ve metanol:su (1:1) karışımı ile ekstrakte edilmiştir. En yüksek antioksidan aktivite ve toplam fenolik içeriği metanol:su ekstraktında elde edilmiştir. Prosiyanidin B1 ve B2, (-)-epikateşin, epigallokateşin gallat ve rutin ekstraksiyonunda en iyi çözücünün metanol:su karışımı olduğu, gallik asidin suda daha iyi ekstrakte edildiği tespit edilmiştir.

Abstract

Extraction of bio compounds from plant materials is one of the most important step of the processes such as dietary supplement production and recovery of the compounds from the industrial wastes. It is highly affected from the factors such as solvent type, particle size, extraction time and temperature. To determine the total phenolics, individual phenolic profile and antioxidant activity of Hawthorn (Crataegus orientalis) fruit extracted with different solvents and to specify the best solvent for extraction of phenolics were aimed in this research. Fruits, picked from wild growing trees in Beyşehir, were extracted with water, methanol and methanol:water (1:1) mixture. Highest total phenolics and antioxidant activity were observed in methanol:water extract. While gallic acid was extracted effectively in water, methanol:water mixture was the best solvent for extraction of procyanidin B1, procyanidin B2, (-)- epicatechin, epigallocatechin gallate and rutin.
