Yüksek bor konsantrasyonlarının Triticum durum desf. cv. Gediz' de büyüme ve protein metabolizması üzerine etkileri

Özet
Bu çalışmada, 15 günlük buğday (Triticum durum Desf. ev, Gediz) fidelerinin, sera koşullarında yapılan denemelerde yüksek bor (B) konsantrasyonlarına büyüme ve protein metabolizması açısından verdiği tepkiler irdelenmiştir. Kontrol bitkiler bir ay boyunca Hoagland besin çözeltisi ile ve bor uygulaması yapılan bitkiler ise Hogland besin çözeltisinde, B mikro elementinin miktarı 5, l O, 20 kat (2,3: 4,6; 9,2 mg kg'1 B) artırılarak yetiştirilmiştir. Elde edilen sonuçlara göre, kök ve gövde boyu açısından, kontrole göre 9,2 mg kg'1 B uygulamasında önemli bir düşüş belirlenmiştir. Borun, kök kuru madde üretimindeki engelleyici etkisi, gövde kuru madde üretimine benzer şekilde ortaya çıkmıştır (sırasıyla % 46 ve 52). Bor uygulaması yapılan bitkilerde fotosentetik metabolizma olumsuz yönde etkilenmiş ve yapraklardaki toplam klorofil miktarı düşerken, klorofil a/b oranı kontrole göre artmıştır. Kontrol ve bor uygulanmış bitkilerde tohumlardan elde edilen proteinler, SDS-PAGE tekniği kullanılarak ayrılmış ve Coomassie Brillant Blue boyaması ile görünürleştirilmiştir. Bor stresi tohumlarda toplam protein miktarında önemli değişikliğe neden olurken, tohum proteinlerinin sayısında azalma veya artışa neden olmamıştır. Bu parametreler göz önünde tutularak, "durum " türü buğdaylarda Gediz kültür varyetesinin kor fazlalığına toleranslı olduğu belirlenmiştir.
Abstract
Experiments were carried out to study growth and protein metabolism responses offlfteen-day-old seedlings of wheat (Triticum durum Desf. cv. Gediz) to high boron (B) concentrations in green house conditions. Control plants were grown on Hoagland nutritional solution for one month. Boron treated plants were irrigated same solution except replacing normal boron amount with 5, 10 and 20 fold boron (2.3, 4.6, 9.2 mg kg'1 B). In the respect of root and shoot height, there were significant decrease in 9.2 mg kg'1 B application compared with control. The detrimental effect ofB on root dry matter production was obtained similar degree on shoot dry matter (52 and 46%, respectively). Photosynlhetic metabolism in B treated plants were influenced negatively in the terms of decreasing of total chlorophyll contents and increasing in chlorophyll a/b ratios relative to the control. In seeds there were remarkable changes in total protein content after B treatment, relative to the control. Total protein patterns were obtained by analysis of total protein extract from seeds of control and B-treated plants using SDS-PAGE followed by Coomassie Brillant Blue staining. Coomassie Brillant Blue stained gels showed that B stress caused no increase or decrease in a number of proteins in seeds. These parameters show that Triticum durum Desf. cv. Gediz cultivar is tolerant of high boron concentrations.
