DEVELİ YÖRESİNDE SIĞIR BRUSELLOZUNUN SEROLOJİK TESTLERLE (RBPT, SAT, C-ELISA, CFT) TEŞHİSİ

Özet
Bu çalışma, Kayseri ili Develi ilçesinde abort yapan sığırların serumlarında 4 farklı serolojik test kullanılarak Brucella abortus karşı şekillenen antikorların ortaya konulması; kullanılan serolojik testlerin teşhisteki duyarlılıklarının saptanması ve bölgede sığır brusellozun görülme gerçekleştirilmiştir.Develi ilçesinde abort yapan 200 baş sığırdan kan örnekleri toplanmıştır. Serumlar Rose Bengal Plate Test (RBPT)’i, Serum Aglütinasyon Test (SAT)’i, Kompetatif ELISA (C-ELISA) ve Komplement Fikzasyon Test (CFT)’ine tabi tutulmuş, testlerin duyarlılıklarının değerlendirilmesinde CFT standart test olarak kabul edilmiştir.İncelenen 200 serumunun 57 (% 28,5)’si RBPT ile pozitif bulunurken geri kalanın negatif olduğu saptandı. Serum aglütinasyon test ile serumların 22 (% 11)’sinin pozitif, 168 (% 84)’inin negatif ve 10 (% 5)’unun şüpheli olduğu belirlenirken C-ELISA ile 36 (% 18)’sı pozitif ve 164 (% 82)’ü negatif olarak tespit edildi. Diğer taraftan CFT ile 30 (% 15)’u pozitif, 163 (% 81,5)’ü negatif ve 7 (% 3,5)’si şüpheli olarak değerlendirildi. Brusellozun teşhisinde her üç testinde duyarlı oldukları, CFT ile C-ELISA arasında diğer testlere göre daha iyi bir uyumluluğun bulunduğu saptandı. Develi ilçesinde atık yapan sığırlarda brusellozis görülme oranının % 15 olduğu belirlendi.Sonuç olarak, Develi yöresinde sığırlarda atık olgularının yüksek bir görülme oranına sahip olduğu ve halk sağlığı açısından da büyük bir risk oluşturabileceği belirlenmiştir. Kısa sürede ve güvenilir sonuçlar alındığından sığır brusellozunun serolojik teşhisinde CELISA’nın daha üstün olduğu kanaatine varılmıştır
Abstract
This study was carried out to detect the antibodies against Brucella abortus in the sera of aborted cows reared in Develi district of Kayseri province with four different serological tests and to compare the sensitivity of the applied serological tests as well as to determine the incidence of bovine brucellosis in the region. Blood samples were collected from the 200 aborted cows in Develi district. Sera were tested with Rose Bengal Plate Test (RBPT), Serum Agglutination Test (SAT), Competetive ELISA (C-ELISA) and Complement Fixation Test (CFT) and the CFT was accepted as the standard test for the evaluation of the sensitivity of the tests. Of 57 (28.50 %) out of 200 tested sera were found to be positive with RBPT and the remaining sera were found as negative. Of 22 sera (11 %) were found to be positive, 168 (84 %) of the sera were found to be negative and 10 of the sera were found to be suspicious with SAT whereas 36 (18 %) of the sera were positive and 164 (82 %) of the sera were negative with CELISA. On the other hand, CFT revealed that 30 (15 %) of the sera were positive, 163 (81.50 %) of the sera were negative and 7 (3.50 %) of the sera were suspicious. For the diagnosis of brucellosis, all of the tests were found to be sensitive, the results of C-ELISA were found to be more consistent with the results of CFT than the other tests. The frequency of brucellosis in aborted cows in Develi district was determined as 15 % according to the results of the tests. In conclusion, it has been determined that the incidence of abortus in cattle in Develi district is high, and it may pose a high risk for public health. Because rapid and reliable results were obtained with the C-ELISA, this test is considered to be superior to other investigated tests for the serological diagnosis of bovine brucellosis.
