Erişkin ve fetal hepatositlerin ''in vivo'' ve ''in vitro'' yapısal ve fonksiyonel özelliklerinin değerlendirilmesi

Özet
Amaç: Çalışmamızda, erişkin ve fetal hepatositlerinin in vitro fizyolojik ve morfolojik özelliklerini ve taşıyıcı poli-mer üzerinde omentuma implantasyon sonrasında elde edilmiş yeni karaciğer dokusunun yapısını incelemeyi amaçladık. Gereç ve Yöntem: Karaciğer doku örnekleri erişkin ve 20. gestasyon günündeki fetal Yeni Zelanda tipi tavşanlardan alındı. Dokular kollajenaz ile parçalanarak hepatositler elde edildi. Hücre süspansiyonu kollajenle kaplı flasklara ekildi ve hepatosit besiyeri (H 1777, Sigma) kullanılarak standart kültür şartlarında üretildi. Flask yüzeyini dolduran hücrelerin morfolojik özelliklen boyanmadan, ışık mikroskobu kullanılarak değerlendirildi. Hepatositler tripsin-EDTA ile kaldırıldı ve hücre çözeltisi sayılarak kollajenle kaplı poliglikolaktik asit (PLGA) polimerleri üzerine her birine yaklaşık "4x106" hücre olacak şekilde ekildi. Hücre-polimer kompleksi 10 gün boyunca kültüre edildi, erişkin hepatositler otolog, fetal hepatositler homolog omentum içine implate edildi ve non-absorbable sutürle tespit edildi. Üç hafta sonra implantlar rezeke edildf ve parafine gömüldü. Bloklardan alınan kesitler hemotoksileneozin, PAS ve best carmine boyaları ile boy andı, ışık mikroskobu ile histolojik özellikleri incelendi. Bulgular: Erişkin ve fetal,, hepatositler başarılı biçimde izole edilerek üretilmiştir. Üreyen hücrelerin hemen tümü¬nün hegzogonal şekilli, büyük santral yerleşimli nükleuslu olduğu ve büyük kısmının çift nükleus içerdiği görüldü. İmplantın parafin bloktan elde edilen kesitlerinde dokunun iyi organize olduğu ve doku içinde vasküler yapıların oluştuğu gösterilmiştir. Hepatositlerin intakt polimer lifleri arasında kümeler oluşturduğu gösterilmiştir. Sonuç: Çalışmamızda hepatositlerin uygun taşıyıcı polimer üzerinde taşınarak implate edildikleri doku içinde kitle oluşturmak üzere üremelerine devam ettikleri görülmüştür. Fetal ve yetişkin karaciğer hücrelerinin morfolojik incelemesinde fark bulunmamakla birlikte, fetal hücrelerin daha hızlı çoğaldıkları görülmüştür. Kollajen kaplı PLGA polimerlerinin karaciğer hücreleri için iyi bir taşıyıcı iskelet olduğu görülmüştür. Omentumun, implantın adaptasyonu ve vaskülarizasyonu için iyi bir yapı olduğu görülmüştür. Bu modelin, yapay karaciğer dokusu elde ederek ortotopik transplantasyona hazırlamak için uygun olduğu düşünülmektedir.
Abstract
Aim: In this study, we aimed to examine the in vitro physiological and morphological characteristics of fetal and adult hepatocytes and in situ tissue masses after implantation into the omentum. Materials and Methods: Liver tissue samples were obtained from fetuses of twenty-day-gestation age and adult New Zealand rabbits. Hepatocytes were separated by collagenase digestion. Cell suspension was seeded onto collagen coated dishes and cultivation was maintained using hepatocyte medium (H1777, Sigma) under standard culture condition. Growing cells were examined morphologically using light and electron microscopic images. After reaching confluence, flasks were treated with trypsin-ED-TA and cell suspension was seeded onto the collagen coated unwoven poliglikolactic acid (PLGA) polymers and cultivated for 10 days. Fetus and adult cell-polymer complexes were implanted (fixed with sutures) into the omentum of rabbits homologously and autologously respectively. After three weeks of implantation, tissue masses were resected and embedded into paraffin. Paraffin sections were stained with hemotoxilen-eosine, PAS, best carmine and examined histologically. Results: Both fetal and adult hepatocytes were successfully isolated and cultivated. Morphologic analyzes have shown that all cell populations expressed typical spheroid shape with centrally located nucleus. Fetal cells reached conflu¬ence in two weeks whereas it took three weeks for the adult liver cells. Attached and proliferated hepatocytes on the polymer fibers were detected after 24 hours of seeding onto the scaffolds. Histological examination of artificial tissues harvested from omentum has shown well organized and vascularized liver tissues. Conclusion: Although fetal liver cells seem proliferating rapidly than adults, there is no difference in histological examination of in situ tissue masses. Collagen coated PLGA polymers may be the preferred vehicle for cell transfer. We think that omentum is the most suitable tissue for adaptation and vascularization of hepatic cells. This model can be used to obtain artificial hepatic tissue and also for orthotopic transplantation.
