Özet

Amaç: Seramik materyaller biyolojik, mekanik ve estetik gibi pek çok yararlı özelliği sayesinde Diş Hekimliği pratiğinde yaygın olarak kullanılır. Ancak yapısı ve yüzey karakteristiği değiştikçe klinik sonuçlar etkilenebilir.Metod: Feldspatik seramikten (n= 50), lityum-disilikat bazlı seramikten (n= 50) ve yitrium-stabilize zirkonya seramikten (n= 50) 0.5 mm kalınlıkta ve 2 mm yarıçaplı disk şeklinde örnekler hazırlandı. Bu çalışmada, biyofilm değerlendirmesi için 4 farklı mikroorganizma seçildi. Sitotoksisite testi için fare fibroblast hücreleri (McCoy cells) kullanıldı. Yüzeye yapışan mikroorganizma boyanmalarının optik yoğunluğu mikroeliza ile 620 nm dalgaboyunda spektrofotometrik olarak ölçüldü. Tek yönlü ANOVA, Post-hoc testleri, bağımsız T testleri, kruskall-Wallis ve Pearson Korelasyon testi kullanılarak istatistiksel analizler yapıldı.Bulgular: Lityum-disilikat (IPS) bazlı seramik grubunda bulunan hücreler 24 saat sonra en yüksek yaşam değerlerini gösterdi. Feldspatik grup en düşük optik yoğunluk değerleri ortaya çıkardı. Pearson korelasyon testi 24 ve 48 saat sonra biyofilm oluşumu ve fibroblast yaşam değerleri arasında doğrusal bir korelasyon göstermedi.Sonuç: Bu çalışmada kullanılan bütün dental materyaller sitotoksik reaksiyon ortaya çıkardı.

Abstract

Background and Aim: Ceramic materials are widely used in dental practice because of their many beneficial properties like biological and mechanical features and aesthetics. However as the structure and surface characteristics of these restorations change the clinical outcomes may be altered.Materials and methods: Disc shaped dental ceramic specimens with 0.5 mm thickness and 2 mm radius were fabricated from feldspathic ceramic (n=50), lithium di-silicate based ceramic (n=50) and yttrium stabilized zirconia based ceramic (n=50). Four different microorganisms were selected for this study for biofilm formation evaluation. Mouse fibroblast cells (McCoy cells) were used for the in vitro cytotoxicity tests. Optical density (OD) of stained adherent bacteria was determined with a micro ELISA auto reader at wavelength of 620 nm spectrophotometrically. One-way ANOVA, Post Hoc tests, Independent Sample-T tests, Kruskall-Wallis test and the Pearson Correlation tests were used for statistical analysis.Results: The survival of fibroblast cells at Lithium di-silicate based ceramic group (IPS) was the highest after 24 hours. Feldspathic group exhibited the lowest mean optical density after 48 hours. The Pearson Correlation test did not reveal a linear correlation between biofilm formation and fibroblast survival after both 24 and 48 hours.Conclusion: All the dental materials used in this study induced cytotoxic reactions.
