BIOFILM QUANTIFICATION FORMED ON 3 DIFFERENT ADHESIVE RESIN MATERIAL

Özet

Background and Aim: Dental adhesive materials should bond to both dentine and restorative material quite well and should act as a protective barrier/plug for the interface. They may prone to form biofilms on their surfaces and may cause many diseases, from periodontal problems to systemic infections. The aim of this study is to evaluate the biofilm forming potential of 3 different microorganisms, which can be colonized quickly on 3 different dental adhesive materials, by measuring the optical density quantitatively. Materials and Methods: Disc-shaped samples with 1 cm diameter and 1 mm depth were prepared from 3 different adhesive material. Strains of S. aureus ATCC 29213, S. mutans ATCC 25175 and C. albicans ATCC 90028 were selected to form biofilms on the material surfaces. The optical density of the samples was measured with a micro ELISA reader with a spectrophotometric method at 620 nm wavelength. Results were evaluated using Welch-ANOVA and Kruskall-Wallis parametric and non-parametric tests. Results: S. aureus shows the highest biofilm formation on PanaviaF 2.0 samples when all subgroups were compared. The least biofilm formation was observed on Maxcem Elite samples with C .albicans (0.0811±0.0009 mg/µl). Considering the first triple-subgroup, the optical density was the highest (0.1289±0.0090 mg/µl) on PanaviaF 2.0 samples. Considering each microorganism (second triple sub-group), S. aureus formed the highest optical density value (0.1100±0.0205 mg/µl). Conclusion: Dental adhesive materials used for cementation have a structure including hallow spaces and microorganisms at the interface. Therefore, biocompatibility is still a matter of debate

Abstract

Amaç: Dental adeziv malzemeler hem dentine hem de restoratif materyale oldukça iyi bağlanmalı ve arayüz için koruyucu bir bariyer/tıkaç görevi görmelidir. Bu materyaller yüzeylerinde biyofilm oluşturmaya eğilimli olabilirler ve periodontal problemlerden sistemik enfeksiyonlara kadar birçok hastalığa neden olabilirler. Bu çalışmanın amacı, optik yoğunluğu kantitatif olarak ölçerek, 3 farklı dental yapışkan materyal üzerinde hızlı bir şekilde kolonize edilebilen 3 farklı mikroorganizmanın biyofilm oluşturma potansiyelini değerlendirmektir. Gereç ve Yöntem: 1 cm çapında ve 1 mm derinliğinde disk şeklindeki numuneler 3 farklı adeziv malzemeden hazırlanmıştır. Malzeme yüzeylerinde biyofilm oluşturmak üzere S. aureus ATCC 29213, S. mutans ATCC 25175 ve C. albicans ATCC 90028 suşları seçilmiştir. Numunelerin optik yoğunluğu, 620 nm dalga boyunda spektrofotometrik yöntem ile bir mikro ELISA okuyucu ile ölçülmüştür. Sonuçlar parametrik ve non-parametrik Welch-ANOVA ve Kruskal-Wallis testleriyle incelendi. Bulgular: S. aureus, tüm alt gruplar karşılaştırıldığında PanaviaF 2.0 numunelerinde en yüksek biyofilm oluşumunu göstermektedir. En az biyofilm oluşumu C. albicans (0.0811 ± 0.0009 mg / µl) tarafından Maxcem Elite numunelerinde oluşturulmuştur. İlk üçlü alt grup göz önüne alındığında, optik yoğunluk PanaviaF 2.0 numunelerinde en yüksek (0.1289 ± 0.0090 mg / µl) bulunmuştur. Her bir mikroorganizma göz önüne alındığında (ikinci üçlü alt grup), S. aureus en yüksek optik yoğunluk değerini (0.1100 ± 0.0205 mg / µl) oluşturmuştur. Sonuç: Simantasyon amacıyla kullanılan dental adeziv malzemelerin ara yüzde oluşturduğu tabaka mikroorganizma içeren boşluklu bir yapıya sahiptir. Bu nedenle biyouyumluluk özelliği halen tartışma konusudur
